چکیده

Background Leptospirosis is an emergent zoonotic disease caused by pathogenic Leptospira spp., and humans are regarded as an accidental or dead-end host [1]. The global incidence of severe human leptospirosis was estimated to be approximately 500.000 cases worldwide, although this is likely an underestimate due to the lack of a reliable diagnostic test. Mortality ranges from 10 to > 50% for Weil’s disease or severe pulmonary hemorrhage syndrome (SPHS), respectively [2]. The microscopic agglutination test (MAT) is the standard test for this disease, however, it requires paired serum samples, is laborious, difficult to analyze and subjective [3]. Apart from the danger connected with rodents, which are the main vectors of leptospires, occurrence of the disease in dogs can generate a higher risk of infection for humans. Thus, a novel sensitive and specific serological test providing a rapid and secure diagnosis is urgently required for the laboratorial diagnosis of leptospirosis. In this study, 15 recombinant proteins from L. interrogans were expressed, purified and evaluated in an indirect ELISA using canine sera that were characterized by the MAT and a whole-cell Leptospira ELISA.

Cloning and Expression of Leptospira LipL32 Antigen as a Candidate for Rapid Diagnosis

Background and Objective: Leptospirosis as an important emerging infectious zoonotic disease caused by spirochetes of the genus Leptospira. Given the low sensitivity and long duration of its culture, the diagnosis of leptospirosis is mainly based on serological methods. The microscopic agglutination test (MAT) is considered as the reference method. Because of the complexity of the MAT, there is...

Development of a recombinant protein based dot-blot for the diagnosis of canine leptospirosis

Background Leptospirosis is a globally disseminated zoonosis, recognized as a re-emergent neglected disease [1] and the causative pathogens are spirochetes from genus Leptospira [2]. In Brazil, more than 100.00 leptospirosis cases are reported annually, with outbreaks emerging especially during floods [3]. The microscopic agglutination test (MAT) is considered the standard serological test for ...

Expansion of a highly sensitive and specific ELISA test for diagnosis of hydatidosis using recombinant EgB8/2 protein

Objective(s): Hydatidosis is a zoonotic infection and endemic in Iran. Due to the serological cross-reactivity (of sera) with other parasitic infection, diagnosis of hydatid cyst is considered to be problematic. In this regard, application of recombinant antigens improves serological diagnosis for human hydatidosis. Here, we present an ELISA test based on B8/2 recombinant antigen of Echinococcu...

Recombinant Antigens rLipL21, rLoa22, rLipL32 and rLigACon4-8 for Serological Diagnosis of Leptospirosis by Enzyme-Linked Immunosorbent Assays in Dogs

Animal leptospirosis is one of the most common zoonotic diseases in the United States and around the world. In a previous study, we applied four recombinant antigens, rLipL21, rLoa22, rLipL32 and rLigACon4-8 of Leptospira interrogans (L. interrogans) for the serological diagnosis of equine leptospirosis (Ye et al, Serodiagnosis of equine leptospirosis by ELISA using four recombinant protein mar...

Development of New Recombinant DgK Antigen for Diagnosis of Dirofilaria immitis Infections in Dogs Using ELISA Technique and Its Comparison to Molecular Methods

Background: Dirofilaria immitis is a cosmopolitan zoonotic, vector-borne parasite of carnivorous animals causing dirofilariasis in human beings. Common commercial serodiagnostic tests for canine dirofilariasis usually lead to different results in their sensitivity and specificity. The present study reports development of recombinant DgK (rDgK) antigen of D. immitis for accurate immunodiagnosis o...
